Best Substrate To Start Tissue Culture

    For awhile now I’ve been using as powder in 9oz cups to start tissue culture plants in my high tech 40b farm tank. My reasoning behind the cups is to have “mother” plants to take cuttings from for my other projects. After having a few species melt I’m starting to question of using the as powder is best, or, if perhaps the soil is a bit “hot” for the youngster and if I should try using an inert substrate eg ec, or similar. The species that have melted have been Lud super red, P hellferi, E Hydropiper, Ammania Sulawesi, and mermaid weed.
    Any advice and or thoughts on this would be greatly appreciated.
